Summary of thesis | The occurrence of vasospasm following subarachnoid hemorrhage (SAH) induces early intracellular changes leading to the release of specific proteins in the brain extracellular fluid.
The aim of this project is to characterize the proteome of brain extracellular fluid (ECF) (microdialysis sample) of 2 groups of SAH patients, presenting or not delayed cerebral ischemia (DCI) and to highlight proteins differentially expressed.
Microdialysis protein content will be identified, quantified and compared using quantitative mass spectrometry. Among the proteins differentially expressed, 8-10 most promising proteins will be selected (siginificant down or overexpression, cellular location and function in relation to the context of the project) and verified. Finally the clinical performances of 2-3 DCI biomarkers will be then validated by classical immunoassays on the blood of independent SAH cohorts.
This project is a unique opportunity to highlight new blood vasospasm predictors allowing clinicians to better manage SAH patients and therefore potentially reduce the tragic consequences associated to vasospasm. |
