Description |
The course will focus on light microscopy, and the objective of the course is two-fold: 1) to acquire or strengthen basic knowledge of the most common methods of plant optical imaging (including sample preparation and staining); 2) to acquire or strengthen basic skills in image acquisition and in image analysis. The course will include two lectures on basic and advanced fluorescence microscopy and sample preparation. In parallel, various practical sessions will be held: • hands-on sessions on fluorescence microscopes; • a hands-on session on image analysis; • an introductory session on plant histology (sectioning with a microtome and a vibratome, staining and mounting). For the microscopy and histology practical sessions, participants will be grouped by 3 and the groups will rotate between different setups. These sessions will be done with samples brought by the participants. During the image analysis hands-on, participants will work on the software Fiji; this will be done on the participants' personal laptops. During the microscopy sessions, all participants will use three different setups, including widefield as well as CLSM confocal microscopes.
The participants are expected to have prior experience with microscopy (and preferably with confocal microscopy). Participants will be asked to bring their own laptops for the image analysis hand-on session.
The practicals will take place at the Bioimaging Center of UniGE.
